I'm trying to get the third proficiency thing on the second row (shown circled below). I have the 2 points it says it costs, so why won't it let me buy that ability? Do I need to buy all five levels of the one before it first?

Yup, you'll need to put 5 points in the first bolt in that row to get to the next upgrade after it.
